TCR? chain repertoire of peripheral ?? T cells is generated through the step-wise assembly and subsequent selection of TCR? variable region exons during thymocyte development. To evaluate the influence of a two-step recombination process on V? rearrangement and selection, we generated mice with a pre-assembled D?1J?1.1 complex on the J?1? allele, an endogenous TCR? allele that lacks the D?2-J?2 cluster, creating the J?1DJ? allele. As compared to J?1?/? mice, both J?1DJ?/? and J?1DJ?/DJ? mice exhibited grossly normal thymocyte development and TCR? allelic exclusion. In addition, V? rearrangements on J?1DJ? alleles occurred at the same frequency as V? rearrangements on J?1? alleles, and were similarly regulated by TCR? mediated feedback regulation. However, thymocytes with V?DJ? rearrangements assembled on J?1DJ? alleles overall were preferentially selected and the V? repertoire of ?? T cells was significantly altered during ?? TCR selection in J?1DJ?/? and J?1DJ?/DJ? mice, as compared to in J?1?/? mice. Our data indicate that the diversity of DJ? complexes assembled during thymocyte development influences TCR? chain selection and peripheral V? repertoire.
