  • Using gene editing, the authors have generated human cell lines lacking Dicer and found that these cells are essentially devoid of microRNAs. However, transfected synthetic microRNA duplexes rescue microRNA function and permit the identification of the mRNA targets of single microRNAs in the absence of endogenous microRNAs. The authors also show that the Drosophila Dicer1 protein, together with its cofactor Loqs-PB, is able to rescue human pre-microRNA processing, indicating strong functional conservation.
