These studies focused on how macrophage activation induced through Toll-like receptor 4 (TLR4) modulates the expression of pro- and anti-inflammatory cytokines. Using RIP-Chip analysis, the authors identified mRNAs that are differentially associated with heterogeneous nuclear ribonucleoprotein K (hnRNP K), a known mRNA-specific translational regulator in differentiating hematopoetic cells. One of these targets, transforming growth factor-?-activated kinase 1 (TAK1), is indeed bound by hnRNP K via a sequence in the 3? UTR in uninduced cells. However, macrophage activation apparently triggers the release of hnRNP K, thus relieving translational repression. The result is that newly synthesized TAK1 can promote an appropriate inflammatory response.
